Custom Stable Cell Lines Using our lentiviral technology, we can generate several types of stable cell lines using GFP, Luciferase, or other markers, and optionally use tet-inducible expression. The service covers some "exotic" cell lines as well as the more common cell lines. Cells that are difficult to grow, such as primary cells, will require additional cost and time. We typically deliver uncloned populations of cells in 1.5-2 months at prices ranging from $10,000 to $20,000, and clonal cell lines in 3-4 months at prices ranging from $20,000 to $30,000. In comparison, other lentiviral stable cell line service providers charge up to $80,000 per cell line. The benefits of using lentiviral technology over traditional methods for stable cell line generation are listed below.
Inducible Pluripotent Stem (iPS) cell lines NEW Cellecta now offers a custom iPS cell line generation service to provide researchers with a novel alternative to embryonic stem cells. By introducing combinations of 4 transcriptional factors to the cells of your choice via lentiviral vectors, we create a clonal population of iPS cells. These cells function as closely as currently possible to ES cells.1,2 Transcriptional factor-dependent Reporter cell lines Used for your high-throughput functional RNAi and phage display screens, and HTS of chemical libraries. The main benefit of using lentiviral technology in generating reporter cell lines is that expression of the reporter gene indicates activation of a given transcriptional response element (TRE) by the cognate transcriptional factor in the natural chromosomal environment rather than in the episomal state in the nucleoplasm as is the case for conventional plasmid-based TR vectors. Tandem copies of integration can be avoided, thus allowing for faithful promoter regulation. To order, you choose the cell line, the transcriptional factor you wish to study, and the reporter gene. Knockdown (shRNA) cell lines Just choose the gene you want to suppress, your cell line, and a reporter gene, and we can produce a stable knockdown cell line. We provide the shRNA design, cloning into the lentiviral vector of your choice, and verification of knockdown. Overexpression (cDNA) cell lines We clone a gene of your choice (up to a 3kb ORF) into a lentiviral vector, and transduce the cell line of your choice to create a stable cell line which overexpresses the gene.
