High Throughput RNAi Functional Screening

Cellecta offers a comprehensive high throughput functional screening service using focused, genome-wide, or custom non-validated or functionally-validated pooled lentiviral shRNA libraries. Below are the options to choose from. We can perform any or all of the experimental steps for you.

Option 1. Customer performs the screen.

Cellecta...

• Provides technical support to help design screening protocol with pooled shRNA library
• Provides a reporter cell line (if necessary), which can be purchased from Cellecta, compatible with the RNAi screen with pooled shRNA library
• Performs data analysis: customer sends send us infected/treated cell samples, and we purify RNA/DNA, amplify shRNA inserts, perform HT sequencing and statistical analysis, and then send the proprietary data back to customer. (Cellecta does not own any rights to data—we perform services under a confidentiality agreement.)

Option 2. Cellecta performs the screen.

Cellecta...

• Provides a custom reporter cell line (if necessary)
• Transduces cells with the pooled shRNA library
• Performs functional testing of cells with a specific phenotype or cellular response
• Purifies RNA/DNA from control and selected samples
• Amplifies shRNA inserts and performs hybridization and/or HT sequencing
• Performs statistical analysis of the digital RNA screening data set

For Option 1, the typical time to completion is 2 months. Prices start at $3,000 per sample, and a minimum of 3 samples (plasmid, control cells, and selected cell fraction) is required. The development of a reporter cell line takes about 2-4 months and is priced at $15,000—25,000 depending on several factors, such as the vector, selection marker, and whether it is a clonal or non-clonal cell line.

Option 2, the complete functional screening service, may take up to 4 months and is priced between $20,000—50,000 depending on the complexity of the RNAi screen. The price will decrease as our level of involvement decreases.

We also offer additional, downstream services:

• Pathway Analysis: We can overlap your data set with the pathways affected by each of the identified siRNA clones
• Digital Gene Expression Profiling Service, from the purified RNA samples
• Confirmation of RNAi screening studies, by performing a second round of RNAi screening with a sub-library generated from the results of the first screen
• Generation of individual shRNA constructs, for validation studies
