Combining pooled lentiviral peptide libraries, FACS sorting of reporter cell lines demonstrating modulation of specific signaling pathways, and HT sequencing for identification of enriched peptides offers a unique, cost-effective alternative to small molecule libraries for drug discovery. The peptide screening service is similar to the RNAi screening. With peptide screening, Cellecta can create libraries expressing thousands of various defined peptide sequences, transduce these into cells, select for a particular phenotype (e.g., cell death), and identify peptides that produce the specific phenotype. The peptide libraries can be constructed based on a particular consensus in a protein, incorporate variations on a natural ligand, or reflect any other rational or random designs.
