Using pooled lentiviral-based shRNA libraries, Cellecta has developed an effective, scalable, high-throughput platform for broad range screening and identification of gene function. RNAi genetic screening makes use of discrete stable suppression of specific genes on a large-scale, allowing for loss-of-function screens in mammalian cell systems. RNAi genetic screens provide an unbiased approach to identify genes that act in specific cellular pathways.

RNAi Screening Services

We will perform a pooled lentiviral-based shRNA screening and deliver the end product whether it is a custom pooled shRNA library, treated cells, sub‑libraries, individual constructs, raw data, and/or analysis.  Full-scale screening order typically involves the following steps:

  1. Custom design and construction of pooled shRNA library
  2. Functional testing of cells for specific phenotype or cellular response
  3. Library transduction and functional testing of cells
  4. Preparation and sequencing of samples from various assay points
  5. Data assembly and analysis

The turnaround time and pricing for screening varies, depending on the specifics (e.g., time points, growth rate of cells, size of library, etc.).  However, a full HT RNAi screening typically takes 4-5 months and costs $70,000 to $90,000 for each cell line screened with a 27K shRNA library.  If you are interested in screening, please contact us and we can discuss scenarios and provide details regarding various options.
