HT RNAi Genetic Screens
Cellecta offers a comprehensive high throughput RNAi genetic screening service using our pre-designed validated or custom non-validated pooled lentiviral bar-coded shRNA libraries. We will perform as much of the screen as you require and deliver the end product whether it be a custom pooled shRNA library, treated cells, sub-libraries, individual constructs, raw data, and/or full bioinformatic and pathway analysis. Option A. Client performs the screen.

- Consults with Client, under Confidentiality Agreement, to determine the best plan of action.
- Designs and constructs one or more Custom Pooled Lentiviral shRNA Libraries targeting client's list(s) of genes or a pre-defined list of genes from our Pre-Designed shRNA Libraries. (2-3 months)
- Provides assistance in designing the optimal HT RNAi screening protocol (if necessary).
- (Optional) Provides a clonal reporter cell line appropriate for Client's RNAi screen. (3-4 months, done in parallel with shRNA library construction)
- (Optional) Performs data analysis. Client sends infected/treated cell samples, and we purify RNA/DNA, amplify shRNA-specific bar-codes, perform HT sequencing and statistical analysis, and send the proprietary data back to Client*. (1-2 months)
Option B. Cellecta performs the screen.

- Consults with Client, under Confidentiality Agreement, to determine the best plan of action.
- Designs and constructs one or more Custom Pooled Lentiviral shRNA Libraries targeting client's list(s) of genes or a pre-defined list of genes from our Pre-Designed shRNA Libraries. (2-3 months)
- (Optional) Develops a clonal reporter cell line appropriate for Client's RNAi screen. (3-4 months, done in parallel with shRNA library construction)
- Transduces Client's target cell line with pooled shRNA library(ies) and performs functional testing of cells with a specific phenotype or cellular response. (1-2 months)
- Performs data analysis. We purify RNA/DNA from infected/treated cell samples, amplify shRNA-specific bar-codes, perform HT sequencing and statistical analysis, and send the proprietary data back to Client*. (1-2 months)
* Cellecta does not own any rights to data—all data and IP are assigned to Client.
For Option A, the time to completion can vary from 2 to 4 months (steps 1-4). An additional 1 to 2 months is required for subsequent data analysis (step 5). The development of a clonal reporter cell line requires approximately 3-4 months and can be generated in parallel with pooled shRNA library construction. Complete sample preparation, HT sequencing, and statistical analysis of data can be performed, and with a minimum of 7 samples is required (two sets of triplicates and a control recommended).
Option B, the complete HT RNAi screening service, may take from 4 to 8 months (depending on options) and is priced depending on the complexity of the RNAi screen. The per-cell-line price will decrease as our level of involvement decreases and the requested number of screens increases.
We also offer additional, downstream services:
- Pathway Analysis, by overlapping your data set with the pathways affected by each of the identified shRNA clones
- Digital Gene Expression Profiling Service, starting from the purified RNA samples
- Confirmation of RNAi screening studies, by performing a second round of RNAi screening with a sub-library generated from the results of the first screen
- Generation of individual shRNA constructs, for validation studies
