Overexpression and Knockdown Cell Lines
Using lentiviral technology and industry-leading experience, we can generate high-quality stable cDNA (overexpression) and shRNA (knockdown) cell lines. Choose any markers, promoters, and cell lines, and either tet-inducible or constitutive shRNA expression for knockdown cell lines. The service covers some "exotic" cell lines as well as the more common cell lines. Cells that are difficult to grow, such as primary cells, will require additional cost and time. The delivery time for non-clonal populations of cells is 1.5-2 months and for clonal cell lines, 3-4 months.
The benefits of using lentiviral technology over traditional methods for stable cell line generation are listed below.
- Nearly any cell type can be used, including cells that are difficult to transfect
- An unlimited number of stable clones can be generated, so you have the freedom to choose the greatest expressing or knockdown cell lines by FACS
- It is the only way to generate overproducer cell lines (for recombinant proteins) with multiple integrated expression constructs of transgene
Overexpression (cDNA) cell lines
Your gene of interest (up to a 3kb ORF) will be cloned into one of Cellecta's cDNA expression vectors expressing the marker of your choice. Your cell line is then transduced, generating a stable cell line which overexpresses the gene. The service includes sequence confirmation and Western blot analysis (or assay for biological activity).

Knockdown (shRNA) cell lines
Choose the target gene, cell line, markers, promoters, and tet-regulatable or constitutive shRNA expression, and we will produce a stable knockdown cell line. We design 10-15 shRNA per gene (, generate individual cell lines, measure knockdown activity, and expand the most promising candidate(s).
