Transcriptional Reporter Cell Lines

Using lentiviral technology, we can generate stable transcriptional reporter cell lines using GFP, RFP, Puro, Bleo, or other readout markers. The service covers some "exotic" cell lines as well as the more common cell lines. Cells that are difficult to grow, such as primary cells, will require additional cost and time. We typically deliver uncloned populations of cells in 1.5-2 months and clonal cell lines in 3-4 months.

The benefits of using lentiviral technology over traditional methods for stable cell line generation are listed below.

  • Expression of the reporter gene indicates transcriptional factor activation in the natural chromosomal environment rather than episomally in the nucleoplasm as is the case for plasmid-based cell lines
  • Almost any cell type can be used, including cells that are difficult to transfect
  • An unlimited number of stable clones can be generated, so you have the freedom to choose the best reporter cell lines by FACS

Transcriptional factor-dependent reporter cell lines can be ideal tools for high-throughput RNAi and peptide library screens, and HTS of chemical libraries. The main benefit of using lentiviral technology in generating reporter cell lines is that expression of the reporter gene indicates activation of a given transcriptional response element (TRE) by the cognate transcriptional factor in the natural chromosomal environment rather than in the episomal state in the nucleoplasm as is the case for conventional plasmid-based TR vectors. Tandem copies of integration can be avoided, thus allowing for faithful promoter regulation.
