The CloneTracker XP barcode constructs are selectable by puromycin or hygromycin and a red fluorescent protein (RFP) marker, so cells containing barcodes will fluoresce red. Additionally, with the pScribe vector, the barcode is expressed in the RNA and thus differences in gene expression are detectable with single-cell RNA-Seq methods.
10 lentiviral constructs with expressed barcodes flanked by common primer sequences to facilitate PCR amplifications. The constructs are available as plasmid DNA or pre-packaged lentiviral particles. After transduction into cells, the barcodes are both stably present in the genomic DNA and expressed in the mRNA on the antibiotic transcript. The barcodes can be amplified from genomic DNA, or detected with RNA-seq. With multiple different constructs, several populations of cells can be labeled and then mixed. In this way, multiple cell populations can be labeled, mixed, growth together, and then the representative numbers of descendant cells from each population can be determined.
The barcode constructs have an RFP fluorescent marker and either puro or hygro selection depending on the kit chosen above.
